Over 40 identified genetic variants contribute to white spotting in the horse. White markings and spotting are under selection for their impact on the economic value of an equine, yet many phenotypes have an unknown genetic basis. Previous studies also demonstrate an interaction between MC1R and ASIP pigmentation loci and white spotting associated with KIT and MITF. We investigated two stallions presenting with a white spotting phenotype of unknown cause. Exon sequencing of the KIT and MITF candidate genes identified a missense variant in KIT (rs1140732842, NC_009146.3:g.79566881T>C, p.T391A) predicted by SIFT and PROVEAN as not tolerated/deleterious. Three independent observers generated an Average Grade of White (AGW) phenotype score for 147 individuals based on photographs. The KIT variant demonstrates a significant QTL association to AGW (p = 3.3 × 10−12). Association with the MC1R Extension locus demonstrated that, although not in LD, MC1R e/e (chestnut) individuals had higher AGW scores than MC1R E/- individuals (p = 3.09 × 10−17). We also report complete linkage of the previously reported KIT W19 allele to this missense variant. We propose to term this variant W34, following the standardized nomenclature for white spotting variants within the equine KIT gene, and report its epistatic interaction with MC1R.

RESUMO

Syringomyelia is a form of myelodysplasia defined by the formation of one or more fluid-filled cavities within the spinal cord that do not communicate with the central canal. The defect may be congenital or acquired. Clinical signs correlate to the segment of spinal cord affected and include pain, paresis, proprioceptive deficits, alterations in sensation, scoliosis, and autonomic dysfunction. This report describes the clinical and pathologic changes in a case of acquired syringomyelia in a 10-year-old American Paint Horse mare. The horse had a six-week history of progressive proprioceptive deficits in all four limbs, bilateral pelvic limb ataxia, and muscle fasciculations that were unresponsive to treatment with stall rest, phenylbutazone, and dexamethasone. Syringomyelia was diagnosed postmortem within cervical, thoracic, and lumbar spinal cord segments. Acquired syringomyelia should be considered among differential diagnoses in adult horses displaying progressive neurologic deficits.

Os objetivos deste estudo foram descrever a frequência cardíaca (FC) e os índices de variabilidade da frequência cardíaca (VFC) materna e fetal no terço final da gestação, bem como descrever a evolução do desenvolvimento do sistema nervoso autônomo durante o período fetal e neonatal. Foram avaliados 20 animais de cada categoria, cujos exames eletrocardiográficos, maternos e fetais, foram realizados aos 15 e sete dias pré-parto. Quanto ao eletrocardiograma neonatal, os momentos avaliados foram ao nascimento até as primeiras 48 horas de vida, e posteriormente, uma vez por semana até os 35 dias de idade. Ocorreram diferenças significativas na frequência cardíaca fetal (FCF) no período avaliado, porém os índices de VFC fetais não se alteraram. Não foram encontradas diferenças significativas nos índices de VFC materna. A média da FCF diminuiu significativamente dos 15 para sete dias do pré-parto (95,6±11,4 bpm; 83,1±12,6, respectivamente), entretanto os índices de VFC fetal não diminuíram. Os resultados obtidos da VFC fetal e neonatal deste estudo, quando comparados aos maternos, indicaram predomínio parassimpático durante a fase fetal e, simpático durante a neonatal, até a terceira e/ou quarta semanas de idade, momento no qual se inicia o equilíbrio entre os dois sistemas.(AU)


The aim of this study were to describe the heart rate (HR) and indexes of maternal and fetal heart rate variability (HRV) in final period of pregnancy, as well as to describe the evolution of the development of autonomic nervous system during fetal and neonatal period. There were 20 animals in each category, whose maternal and fetal electrocardiographic examinations were performed at 15 and 7 days antepartum. Neonatal ECG it was evaluated at birth until the first 48 hours of life, and then once a week up to 35 days. There were significant differences in fetal heart rate (FHR) during this period, but the fetal HRV indexes have not changed. There were no significant differences in the rates of maternal HRV. The mean of fetal HR decreased significantly from 15 to seven antepartum days (95.6±11.4 bpm; 83.1±12.6, respectively), though the fetal HRV indexes have not decreased. The results of fetal and neonatal HRV in the present study, when compared to maternal indicate the parasympathetic dominance during fetal and neonatal sympathetic phase during to the third and/or fourth weeks of age, at which point begins the modulation of the two systems.(AU)

Utilizaram-se 195 cavalos Pampa e um grupo-controle de 41 cavalos da raça Paint, provenientes de plantéis de várias regiões brasileiras, com o objetivo de avaliar a eficiência do teste mediante uso de marcadores bioquímicos: albumina (Al) e proteína de ligação da vitamina D (Gc), para identificação dos possíveis indivíduos homozigotos dominantes para o padrão de pelagem tobiano nos cavalos Pampa. Não foram encontrados genótipos AlBB e GcSS, revelando indício de quebra de ligação gênica entre tais locos e o loco tobiano e a ineficácia do teste bioquímico na detecção dos prováveis indivíduos homozigotos dominantes para o padrão de pelagem tobiano nos cavalos Pampa


One hundred and ninety five Pampa horses and a control group of 41 Paint Horses originated from herds located in different Brazilian regions were used to objective of evaluate the efficiency of the biochemical markers albumin (Al) and vitamin D binding protein (Gc) to identify the probable homozygous dominant for the tobiano coat color pattern in Pampa horses. It was not found any genotype AlBB and GcSS, indicating a possible break of the genetic linkage between these loci and the locus Tobiano, as well as the inefficacy of the biochemical test in the detection of the probable homozygous dominant for the tobiano color pattern in Pampa horses
